Altered brain activity in juvenile myoclonic epilepsy with a monotherapy: a resting-state fMRI study

چکیده
Abstract Background Juvenile myoclonic epilepsy (JME) is the most common syndrome of idiopathic generalized epilepsy. Although resting-state functional magnetic resonance imaging (rs-fMRI) studies have found thalamocortical circuit dysfunction in patients with JME, pathophysiological mechanism JME remains unclear. In this study, we used three complementary parameters rs-fMRI to investigate aberrant brain activity comparison that healthy controls. Methods Rs-fMRI and clinical data were acquired from 49 undergoing monotherapy 44 age- sex-matched After fMRI preprocessing, fractional amplitude low-frequency fluctuation (fALFF), regional homogeneity (ReHo), degree centrality (DC) calculated compared between two groups. Correlation analysis was conducted explore relationship local abnormalities features patients. Results Compared controls, exhibited significantly decreased fALFF, ReHo DC cerebellum, inferior parietal lobe, visual cortex (including fusiform lingual middle occipital gyri), increased right orbitofrontal cortex. patients, there no regions reduced No significant correlation observed or DC, features. Conclusions We demonstrated a wide range abnormal brains including prefrontal cortex, default mode network, cerebellum. The results suggest dysfunctions cerebello-cerebral circuits, which provide clue on potential pathogenesis JME.

Altered Local Spontaneous Brain Activity in Juvenile Myoclonic Epilepsy: A Preliminary Resting-State fMRI Study
Purpose. The purpose of this study was to evaluate the regional synchronization of brain in patients with juvenile myoclonic epilepsy (JME). Methods. Resting-state fMRI data were acquired from twenty-one patients with JME and twenty-two healthy subjects. Regional homogeneity (ReHo) was used to analyze the spontaneous activity in whole brain. Two-sample t-test was performed to detect the ReHo di...
متن کاملLevetiracetam monotherapy in juvenile myoclonic epilepsy
PURPOSE To describe our experience with levetiracetam (LEV) as initial or conversion monotherapy treatment for juvenile myoclonic epilepsy (JME). Valproate, the usual first line agent for JME, has chronic adverse effects, particularly for women of childbearing potential. Since JME requires lifetime treatment, chronic adverse effects of therapy are important consideration. METHODS We reviewed ...
متن کاملAltered baseline brain activity in type 2 diabetes: a resting-state fMRI study.
PURPOSE This study aims to investigate whether altered baseline brain activity exists in type 2 diabetes mellitus (T2DM) patients using resting-state functional magnetic resonance imaging (rs-fMRI) and whether abnormal neural activity in the middle temporal gyrus (MTG) is correlated with cognitive function. METHODS T2DM patients (n=28) were compared with nondiabetic age-, sex-, and education-...
متن کاملAltered functional connectivity in default mode network in absence epilepsy: a resting-state fMRI study.
Dysfunctional default mode network (DMN) has been observed in various mental disorders, including epilepsy (see review Broyd et al. [2009]: Neurosci Biobehav Rev 33:279–296). Because interictal epileptic discharges may affect DMN, resting-state fMRI was used in this study to determine DMN functional connectivity in 14 healthy controls and 12 absence epilepsy patients. To avoid interictal epilep...
